I've used RNA based oligo probes targeted to both genomic 16S DNA and
expressed 16S rRNA, as in situ probes to Legionella spp. labelled with
FITC or FAM,
these give good signals in mixed biofilms (created on glass or steel supports
in chemostats or in a model cooling tower). Other labels used include
TRITC and carboxyfluorescein, allowing differential staining with
mixed riboprobes. Please contact me if you need further information.
