Dear all,
I'm recording cell growth in a flow cell and feel that calibration of the
size measurements by image analysis is important.
Most people use fluorescent latex beads for calibration of epifluorescence
microscopes.
I've tried latex and silica beads with a phase contrast microscope, but
they are phase-bright in contrast to the phase-dark bacteria. Since the
optical properties are so different, I don't think that the edge detection
algorithm will give the same results for bacteria and latex beads of the
same size.
Has anyone an idea of what particles of known size are about as
phase-dark as bacteria?
