Hi Netters,
I really have two questions.
This first is a technical one and the second is a request for opinions more or
less.
First off, I ahve GFP staining from a constrcut I injected into e1282 using
MH86 as my injection marker. The staining is intense and I need to get some
pictures of it obviously. My problem is the staining is COMPLETELY quenched
by NaAzide.
How do I go about getting pictures of this? WHat have other people done in
this instance? Should I mate the array into an unc strain or would that worry
you since you are changing the background even more?
Secondly, when do you believe your GFP pattern and when do you consider it
"background" or ectopic expression? I realize sometimes you may not "expect"
it in odd places, but how do you discern when it is real or not? (I am in the
process of doing mosaics.)
