I recently posted a question about visualizing and counting two different
strains being grown in the same culture: a number of people replied (thanks
to you all) and some suggested that one of the strains could be transformed
with the GFP gene. My new question is, will a strain transgenic with GFP
continue to express the gene indefinitely, or will it disappear over a
number of generations?
Also, I was reminded that two strains living in the same culture will cross
breed (due to males); will vigorous shaking of a liquid culture prevent this
from occurring, or will it merely reduce the frequency with which it occurs
(perhaps to a negligible amount)?
Lastly, if one CAN indefinitely maintain GFP in a strain, where could such
a strain be obtained? (We have a microinjection facility here at UMass, but
not a very advanced one; plus, I have absolutely no experience in this area,
so I thought I'd try and sponge some GFP worms off someone else, if I may.)
