C.briggsae cDNA as probe

Hi!

I would like to know if somebody has ever tried to use C.briggsae cDNA as probe
to clone a C.elegans homologue. I've tried to hybridized a southern blot of
C.elegans DNA with such a cDNA probe and the best I can get is a wide smear (wh
ereas with a C.elegans gene probe, I get a very specific signal). The conditio
ns I used for hybridisation were 5% SDS/0.5MNaPO4 pH 7,2/0.5mM EDTA at 43C.
If one of you have obtained good result with that approach, could you please
send me your conditions? Is there some special thing that I should know about
C.briggsae (codon usage? repetitive sequences? etc?)

Thanks in advance for any help you can provide
