Germ line expression

Hi,
I am interested in looking at the dynamics of the Endoplasmic reticulum
following fertilization in C. elegans embryos. In order to visualize it I
need a very probe specific for this organelle-I have a constrct with GFP,
followed by the ER retention sequence "KDEL" My current strategy has been
to make mRNA in vitro and inject it.
I am wondering if it is possible to get germline expression of GFP by
injecting the plasmid. From what I gather most such injected DNA
typically ends up in a extrachromosomal array that can be stably
inherited; however, germline expression is not seen. If I go for a direct
integration (even though it is very rare) would I be able to get germline
exprssion? What kind of a promoter would I need to use?
Would appreciate any feedback. Thanks in advance.

Yours sincerely,

Ananth Badrinath
