A question or two:
What is the best way to perform minipreps on worms? Will a
regular commercial kit (e.g., Promega, Qiagen) do the trick? If so,
about how many plates of worms are sufficient per prep? Also, what's the
best way to ensure that you've washed away all the bacterial worm food?
We will soon need to take frequent sample aliquots from large liquid
worm cultures, for counting later. Can we put these aliquots at 4 degrees
for any length of time without worrying about the numbers of each stage
changing? How long can the worms survive at this temperature, and will they
continue to develop? (Life would be much easier if we didn't have to count
the worms right at the time of sampling; the sampled worms need not remain
alive, they only need to be `frozen' in time).
