Hello,
I apologize if this has been discussed before. I have had no success using
UV irradiation to obtain integrated lines (both rol-6D and
unc-119-rescued), although others here have had some luck. I have
previously used gamma rays with good, consistent results, but as there are
no convenient X-ray or gamma sources at UCSB, UV is my only option now.
For those that have tried UV to obtain integrants, can you send me a reply
stating (a) which machine was used (e.g. Stratagene 'UV Stratalinker
1800'), (b) the energy (dose), and (c) the frequency of integrants per F1?
I would also like to know if anyone has tried using coinjection with
oligonucleotides as described by Craig Mello, and whether this was
successful.
