Rescue Experiments!

I would appreciate if you help me with some valuable suggestions
regarding rescue experiments. I started already with the whole cosmid
and later plan to inject fragments of the cosmid to narrow down and
assess the boundaries of the rescuing fragments.
1)What concentrations of the cosmid and the rol-6 genes should I inject
into the mutant background?
2)What  percentage of F1 rollers can I expect?
3) Is it possible to chase rollers from F2 and subsequent generations? I
ask this since I did not get any F2 rollers, and for your information my
F1 rollers were too less.
4) How many generations is it necessary to study to believe that rescue
is really working?
