About deletion mutant screening PCR

Hi, everybody,

I have studied C. elegans muscle myogenesis.
In our lab, we are conducting deletion mutant screening.
But, we have some problems about lysing mutant pools.
We collected the mutants worms with M9 buffer + streptomycin(100 ug/ml).
After that, in nested PCR, we failed to get any band, only smear pattern.
We thought that the prep. with M9 buffer, not DW is deleterious.
Is right our thought?
We wonder whether M9 buffer prevents the worm lysis.
