Anyway, found a pretty straight-forward protocol for Sudan Black staining in:
The C. elegans PTEN Homolog, DAF-18, Acts in the Insulin Receptor-like
Metabolic Signaling Pathway by Ogg & Ruvkun. Molecular Cell, Vol 2 (Dec.
1998), page 887-893. Specific protocol on page 892.
I was getting dense staining in gravid adults with some minor variability.
I was just washing the worms off the plates into 15 mL tubes or picking
specific worms into 1.7 mL tubes , depending on how precise I wanted to
be, and using a pap-pen for slide visualization. I also washed the worms
4-5 times with M9 after the staining. Hope this helps.
