[Celegans] Staining eggs

Dear All

 

I am looking for a DNA stain that will stain worms, or more specifically
their eggs, so I can then use imaging software to rapidly count the eggs
in utero. Currently we have tried staining with PI with and without
fixation (not bad, not great), YoPro, Draq5 (little staining), Vibrant
Orange DNA dye (all over the place), PHK26 for membranes (unimpressive).
Since none of these seem to be working really well we would welcome any
suggestions. Do we maybe need to make the eggs/worm permeable first? I
should also add that the worms that we are trying to stain are the
nematodes Heligmosomoides polygyrus and Syphacia spp., but I am
presuming that the methods used for C. elegans will work for all
nematodes.
