GFP mRNA expression

Subject: GFP mRNA expression

Does anyone have any experience of obtaining expression from mRNAs coding
for dual GFP fusions of approximately 2 kilobases with extremely high GC
content? I am microinjecting into cells which normally express other GFP
fusion mRNAs very efficiently. Injection of these longer dual GFP mRNAs has
so far only resulted in expression in 1 of 24 cells injected. It seems that
the most likely explanation is problems with secondary structure formation
- does anyone know of a good way to avoid this?
