DNA from endosperm

DNA extraction from endosperm is problematic because of the starch and
polysaccharide content that results in a gelatinous pellet from which DNA or
RNA is difficult to purify.  We have used a modification of the Urea-based
Plant DNA miniprep (The Maize Handbook, pg. 527) to extract DNA from
kernels.  Substitute 0.35M Na2SO4 for the NaCl in the extraction buffer.
BTW, the urea concentration of the extraction buffer is not listed in the
article -- it is 42 g/100 ml urea (ultrapure).
