I am going to study an anticoagulant
in tick saliva. I am going to use PT and
APTT for screening. The end points for
these tests are fibrin formation. My
boss thinks the manual method (pulling
fibrin on the hook) is too subjective.
Therefore I am trying to do it in other
ways, not with fibrometer or coagulation
analyzer, possibly with a spectrophotometer.
I worked with 0.15 ml cuvette and kinetic
program in spectrophotometer. However,
the clot formed will not evenly distribute
inside the cuvette and I am sure some
light could go through the cuvette without
passing through the clot. Also the density of
PT/APTT reagents is not quite different
from that of fibrin.
I am looking forward to answers to solve
the problems or to do the coagulation
tests by other simple objective methods
other than manual method.
