I am seeking advice on how to prepare larvae of
intestinal nematodes from field-collected samples in a
suitable state for PCR assays.
I want to perform PCR assays to identify two species
of nematodes from abomasal samples of reindeer.
Currently, abomasa are frozen soon after dissection in the
field in Svalbard. Larvae are extracted in the laboratory
back in Scotland by classic pepsin-HCl digestion of
mucosa.
Has anyone succeeded in obtaining success in PCR
using such prefrozen material extracted by pepsin-HCl
digestion? Is there a proven method for preparation
of larval samples for PCR from either fresh or frozen mucosa?
