Dear Naouel,

Without knowing more specifics on what exactly 
you are doing and what sort of cultures you are 
trying to obtain it is difficult to advise you on how 
to proceed. It is also not clear what you mean by 
contamination - the Locke's-Egg medium and 
Robinson's medium are intended for xenic culture 
so there must be bacteria present for the amoebae 
to grow. I may be able to help but I need more information!
