I am trying to culture barley mesophyll protoplasts for transient gene
expression studies of Calvin cycle genes.
Unfortunately the viability in culture over 12-20 hours is very variable.
I have tried various culture solutions e.g. Sarhan and Cesar, Sheen, and
Lazzeri. Would a layer of nurse cells (Brome) help?
Any suggestions would be appreciated.
