In microalgae, 90% acetone is the most common solvent, but certain species
such as the chlorophytes and cyanobacteria are problematic. There are several
options, each of which works better under some circumstances: 100% methanol,
acetone with DMSO, dimethylformamide, etc. How you prepare the extract
i.e. grinding in solvent with a tissue homogenizer, extracting liquid N2
freeze/thawed cells, or sonication ia also an important factor.
You should also be aware of possible degradation of chla, which can occur
quite rapidly in some species.
