Maize chloroplast isolation

 my
research about the effects of some triazine herbicides in the z-scheme of th=
e
photosynthetic process. The plant is maize or corn (Zea mays L.). I try to
measure the chloroplastic activities with the ACMA probe in this plant.
Sometimes I have a good quenching, sometimes not. Can you help me to find a
suitable isolation (grinding) and buffer mediums to this C4 plant? What are =
the
differences in the isolation process of maize mesophyll chloroplasts and the
bundle sheath chloroplasts of this "instable" plant? Thank you all in
