I have been using cauliflower as a source of mitos for succinate DH assay.
They work very well. However, I've also tried to assay the leftover
microsomes in order to demonstrate organelle-specific enzymes. Problem is
that the microsomes have a powerful, succinate-independent reducing
activity that can mask and mimic succinate DH (except of course for being
succinate-independent).
Does anyone know what this reducing activity may be? Or how to inhibit it
or eliminate it?
Alternatively, it could be a very interesting enzyme to look at in its own
right, if I had any idea what it was.
