The American Biology Teacher Volume 51, March 1989, pp. 165-167
describes a protocol for establishment and maintenance of carrot callus
and initiation of somatic embryogenesis from callus. Each stage of the
process uses Murashige and Skoog with Minimal Organics (MSMO) medium
with different levels of 2,4-D added. Callus initiation requires 1 mg /
L of 2,4-D, callus maintenance requires 0.4 mg / L and somatic
embryogenesis is initiated when callus is placed on media lacking 2,4-D.
