We actually have been having pretty good luck with Carolina's carrot
callus initiation medium, which we follow 6 weeks later with carrot
shoot initiation medium. It is necessary to add 30 g/l sugar plus agar
or agargel to these media. As suggested by someone earlier, carrots
that still have the tops are most successful. We aim for the vascular
cambium region when taking explants. The recurring problem has been
contamination, but at least part of the class succeeds. Also, there's
no time to root the shoots before the semester is over.... Some
students get light green fluffy callus, and some get the anthocyanins.
I've not been able to discern a pattern.
