[Plant-education] pigment extraction and TLC

Everyone,

I am trying to use TLC for pigment separation in brown, green, and red
algae. I tried it a couple years ago with the same protocol I use for
spinach (grind in mortar and pestle with acetone, back-extract into
petroleum ether, then run TLC with acetone/petroleum ether--10:1 or
1:10, can't recall).
It wasn't successful. If I remember correctly, it seemed that we made
an emulsion when we tried to back extract, and addition of salt still
did not separate the layers. When we placed the "extract" onto the TLC
plates, it really didn't run well, either.

Does anyone have a good protocol for what I am trying to do?

Thanks,

Doug
