Rosie:
Chilomonas is a very active flagelate which swims vigorously. It will
be very hard to grow them on agar if its too dry for them to swim.
And
otherwise, they will not remain in cohesive colonies.
You probably want a "vital" dye which doesn't kill them. I don't know
of a good one for your purposes. Of course there are numerous
dyes for
staining killed and fixed Chilomonas.
