Expression of viral antigens

Hi netters,

We have a postdoc starting in this lab soon, to work on expression
of viral proteins for use as antigens in diagnostic tests (ultimately!).

Can anyone offer advice on what system we should go for?

I kinda favour the baculovirus system, but what about E.coli systems?
Maybe T7 promoter GST fusion?

Two people who've now left did some work on expression. One used
E.coli & found that human antibodies to E.coli antigens gave a high
background noise when the proteins were used in RIAs; the other
worked with the baculovirus system & didn't get the expected high
protein yields.

If there's sufficient interest I'll summarise & post the replies.

Thanks in advance,

Jon Clewley, Virus Reference Division, Central Public Health
Laboratory, Colindale, London.
