I intend to construct an infectious clone of astrovirus cDNA (RNA virus,
polyadenylated, 10 kb). I have managed to amplify the whole genome in one
long-range PCR but am not quite sure about what kind of plasmid to choose.
Has anyone any ideas about adequat copy number and if any ideal plasmid is
available from manufacturers, or if I have to make my own? if it is better
to transfect cell culture with plasmid DNA containing RNA pol I promotor
(for in vivo transcription), or transfect with RNA derived from in vitro
transcription?
